Research Interests: 

My laboratory studies two aspects of epigenetic modification of the mammalian genome: the interaction between genetic of epigenetic variation in mammals, and the epigenetic remodeling of the genome during meiosis. More specifically, we study the mechanisms by which genetic variants cause phenotypic changes through modification of epigenetic states and gene expression. For this project we use several chromatin and DNA methylation analysis approaches. We have mapped trans-acting genetic factors that cause loss/gain of imprinting of several mammalian genes. We have also identified cis-acting polymorphisms in human and mouse genomes that cause preferential expression of one of the parental alleles (allelic expression) of several genes.

Major epigenetic remodeling of the mammalian genome occurs during meiosis. To better understand the molecular mechanisms underlying this process, we focus on the roles of key proteins involved in meiotic pairing and recombination and meiotic silencing in the establishment of gamete-specific methylation patterns

Recent results:

Our recent analysis of DNA methylation in humans shows that at certain loci, DNA methylation levels vary between females and males and depend on the number of X-chromosomes (X-chromosome dosage). These differences in DNA methylation may be established very early in development and affect predisposition to disease after birth.
